REV-ERB proteins help regulate daily cycles in gene expression, including genes tied to lipid and glucose metabolism. SR9009 binds these receptors and alters their activity in cell and animal experiments. Consequences observed in rodents include changes in skeletal muscle oxidative capacity, blood lipid levels, and exercise performance. The precise chain from receptor occupancy to whole-body effects is still an active area of study. Human responses cannot be assumed from rodent data.
Most published work on SR9009 consists of preclinical studies. It is widely sold as a research chemical, a category that does not imply safety, efficacy, or pharmaceutical-grade quality. Sports anti-doping organizations have listed SR9009 as a prohibited substance, and its presence in an athlete sample can lead to sanctions. Legal status differs by country; in several jurisdictions it is not approved for human consumption and may be treated as an unapproved new drug.
Laboratory studies often administer SR9009 by injection because oral absorption appears poor in rodents. Reported pharmacokinetic properties include rapid metabolism and low systemic exposure after oral dosing. Human pharmacokinetic data are sparse, so absorption, distribution, metabolism, and excretion in people are not well defined. Some research explores related REV-ERB compounds with improved drug-like properties. Regulatory approval for any REV-ERB agonist as a human medicine has not been granted to date.
SR9009 is a synthetic small molecule studied as an agonist of the nuclear receptors REV-ERBα and REV-ERBβ, also called NR1D1 and NR1D2. These receptors help regulate circadian rhythms and metabolic gene programs. The compound was developed for laboratory research, not as an approved therapeutic. Its identity is distinct from steroid hormones and selective androgen receptor modulators. Scientific interest centers on how REV-ERB activation changes gene expression in cells and animal models.

SR9009 is frequently discussed alongside other REV-ERB ligands, including synthetic agonists and natural heme-related molecules. Its selectivity for REV-ERB over related nuclear receptors has been measured in binding and reporter assays, though off-target activity at higher concentrations is possible. The compound is prohibited in sport by the World Anti-Doping Agency, and it is not approved for any medical use in major jurisdictions. Products sold online may be labeled as research chemicals, and their identity and purity are not guaranteed by regulatory review.
SR9009 is a synthetic small molecule that acts on the nuclear receptors REV-ERBα and REV-ERBβ. These receptors are part of the circadian clock machinery and normally repress transcription of certain target genes. In laboratory research, SR9009 is used as a chemical tool to study how REV-ERB activity influences metabolism, inflammation, and daily biological rhythms. The compound is not an approved medicine, and its effects in humans remain largely uncharacterized. It is often described as an investigational agent rather than a therapeutic product.
Quality control for research materials includes identity confirmation by nuclear magnetic resonance and purity assessment by high-performance liquid chromatography. Mass spectrometry provides molecular weight confirmation and can detect related impurities. Purchasers should request a certificate of analysis that lists lot-specific data. Online products advertised for human use often lack such documentation. Distinguishing legitimate research material from mislabeled or contaminated samples is a recurring challenge in independent testing, and independent laboratories may use orthogonal methods to verify identity.
Detection of SR9009 in biological samples usually relies on liquid chromatography coupled to tandem mass spectrometry. This approach separates the compound from matrix components and identifies it by mass transitions. Because SR9009 can undergo metabolism, laboratories often look for both parent drug and specific metabolites. Sample preparation may involve protein precipitation or solid-phase extraction. Method validation examines sensitivity, carryover, and interference from related substances, and reference standards are required for accurate calibration.
Storage recommendations for SR9009 reference material typically specify a freezer at -20 °C or lower, with protection from moisture and light. Repeated freeze-thaw cycles can degrade small molecules and introduce variability. Stock solutions in dimethyl sulfoxide are often aliquoted to avoid repeated handling. Stability studies may examine degradation under heat, humidity, and light exposure. The compound's thiophene and nitro groups can participate in reactions that alter analytical signals over time, so such changes affect quantitative results.
Scientific discussion of SR9009 often separates animal evidence from human anecdote. Rodent studies provide controlled data on endurance, metabolism, and gene expression, but they use specific strains, doses, and treatment durations. Human reports are mostly uncontrolled and cannot establish cause and effect. Open questions include oral bioavailability, tissue distribution, metabolic stability, and long-term effects. Review articles generally call for more rigorous pharmacokinetic and safety research before any clinical use could be considered.
Analytical methods for SR9009 typically rely on liquid chromatography coupled with tandem mass spectrometry. The technique can separate the parent compound from related substances and detect low concentrations in biological matrices. Urine and blood are common samples in anti-doping testing, while in vitro studies may use cell culture media. Rapid metabolism and low expected concentrations make method validation important for reliable identification. Exact metabolite patterns can vary by species and are not fully mapped.
Regulatory treatment of SR9009 reflects its investigational status. The compound has no approved human therapeutic indication, and sports authorities prohibit its use. It appears on anti-doping lists as a non-approved substance or metabolic modulator, depending on the list version. Products sold online as research chemicals are not quality-controlled medicines, so their identity and purity can differ from the label. Such products may also contain unlisted compounds, which complicates both testing and safety assessment.

=== Adenovirus and cytomegalovirus === As of 2014, brincidofovir is in Phase III clinical trials for use in humans against cytomegalovirus and adenovirus. Preliminary safety data from a database of 1000 patients supported progression into later phase trials, Chimerix announced in December 2015 that the Phase III trials for use of the drug in preventing cytomegalovirus infection in stem cell transplant patients had failed, and in February 2016 shut down two other late-stage trials for use of the drug in preventing infection after kidney transplants. Brincidofovir is not yet FDA approved for adenovirus or cytomegalovirus due to lack of efficacy in clinical trials. In a trial of brincidofovir for CMV prophylaxis in stem cell transplant patients, brincidofovir was associated with a 15.5% week 24 all-cause mortality compared with 10.1% among placebo recipients. Additionally brincidofovir was associated with increased serious adverse events (57.1% versus 37.6%) compared with placebo. Brincidofovir was initially offered via an FDA expanded access trial; however as of 9 May 2019, Chimerix discontinued clinical trials of brincidofovir for the treatment of adenovirus and discontinued the expanded access program in 2019.
=== Wastewater treatment === A majority of silver nanoparticles in consumer products go down the drain and are eventually released into sewer systems and reach wastewater treatment plants. Primary screening and grit removal in wastewater treatment does not completely filter out silver nanoparticles, and coagulation treatment may lead to further condensation into wastewater sludge. The secondary wastewater treatment process involves suspended growth systems which allow bacteria to decompose organic matter within the water. Any silver nanoparticles still suspended in the water may collect on these microbes, potentially killing them due to their antimicrobial effects. After passing through both treatment processes, the silver nanoparticles are eventually deposited into the environment. A majority of the submerged portions of wastewater treatment plants are anoxic and rich in sulfur. During the wastewater treatment process, silver nanoparticles either remain the same, are converted into free silver ions, complex with ligands, or agglomerate. Silver nanoparticles can also attach to wastewater biosolids found in both the sludge and the effluent. Silver ions in wastewater are removed efficiently because of their strong complexation with chloride or sulfide. A majority of the silver found in wastewater treatment plant effluent is associated with reduced sulfur as organic thiol groups and inorganic sulfides. Silver nanoparticles also tend to accumulate in activated sludge, and the dominant form of the silver found in sewage sludge is Ag2S.
=== Plot === An unnamed character, only referred as "Candidate Twelve," arrives for work at the Black Mesa Research Facility. The facility has flickering of lights and failing electrical equipment, with tremors shaking it as scientists begin to worry about the experiment. After entering a tram, the "resonance cascade" forces Candidate Twelve to fight for survival as the G-Man observes. After the resonance cascade, Candidate Twelve battles through the facility's sewer system, encountering other survivors along the way. The gargantua, a massive alien organism, stalks the player's movements. The player eventually escapes to the surface in a large freight elevator. After taking it, Candidate Twelve meets a construction worker that has a plan to escape. He sends off Candidate Twelve to look for a security guard needed for a door he's stuck behind. At this time, military convoys can be seen entering Black Mesa. The player nearly escapes with the construction worker, a surviving security guard and scientist, but the construction worker turns out to be dead, impaled by crossbow arrows. The rest are ambushed by the HECU marines and killed. Candidate Twelve escapes and battles the military en route back to the transit hub from the start of the game. Along the way, Candidate Twelve encounters visions projected by a sentient entity that grants visions of the Universal Union, also known as the Combine. The entity appears to be fleeing the powers of the enigmatic G-Man. Candidate Twelve arrives at a climactic confrontation between the Xen aliens and the HECU marines.
=== EU risk assessments === In 2002 the EU Scientific Committee on Veterinary Measures relating to Public Health (SCVPH) claimed that the use of beef growth hormones posed a potential health risk, and in 2003 the EU enacted Directive 2003/74/EC to amend its ban, but the US and Canada objected that the EU had not met WTO standards for scientific risk assessment. The EC claimed that the hormones remain in the tissue, specifically 17-beta estradiol. However, the EC found no clear link to health risks in humans for the other five hormones. The EC reported that high amounts of hormones had been found in areas around dense cattle lots. This increase in hormones in groundwater affected waterways and nearby wild fish. Hormone contamination of North American waterways would not, however, have any health impacts on European consumers.

== Diagnosis == Measuring the level of thyroid-stimulating hormone (TSH), produced by the pituitary gland (which in turn is also regulated by the hypothalamus's TSH-Releasing Hormone) in the blood, is typically the initial test for suspected hyperthyroidism. A low TSH level typically indicates that the pituitary gland is being inhibited or "instructed" by the brain to cut back on stimulating the thyroid gland, having sensed increased levels of T4 and/or T3 in the blood. In rare circumstances, a low TSH indicates primary failure of the pituitary, or temporary inhibition of the pituitary due to another illness (euthyroid sick syndrome), and so checking the T4 and T3 is still clinically useful. Measuring specific antibodies, such as anti-TSH-receptor antibodies in Graves' disease, or anti-thyroid peroxidase in Hashimoto's thyroiditis—a common cause of hypothyroidism—may also contribute to the diagnosis. The diagnosis of hyperthyroidism is confirmed by blood tests that show a decreased thyroid-stimulating hormone (TSH) level and elevated T4 and T3 levels. TSH is a hormone made by the pituitary gland in the brain that tells the thyroid gland how much hormone to make. When there is too much thyroid hormone, the TSH will be low. A radioactive iodine uptake test and thyroid scan together characterize or enable radiologists and doctors to determine the cause of hyperthyroidism. The uptake test uses radioactive iodine injected or taken orally on an empty stomach to measure the amount of iodine absorbed by the thyroid gland.
=== All-cause mortality === Two meta-analyses concluded that as a dietary supplement, vitamin E neither improved nor impaired all-cause mortality. A meta-analysis of long-term clinical trials reported a non-significant 2% increase in all-cause mortality when alpha-tocopherol was the only supplement used. The same journal article reported a statistically significant 3% increase for results when alpha-tocopherol was used in combination with other nutrients (vitamin A, vitamin C, beta-carotene, selenium).

== Interactions == Spironolactone often increases serum potassium levels and can cause hyperkalemia, a very serious condition. Therefore, people using this medication should avoid potassium supplements and salt substitutes containing potassium. Physicians must be careful to monitor potassium levels in both males and females who are taking spironolactone as a diuretic, especially during the first 12 months of use and whenever the dosage is increased. Doctors may also recommend that some patients may be advised to limit dietary consumption of potassium-rich foods. Recent data suggest that both potassium monitoring and dietary restriction of potassium intake are unnecessary in healthy young women taking spironolactone for acne and in healthy, young, gender-diverse individuals taking spironolactone for hormone therapy. Spironolactone together with trimethoprim/sulfamethoxazole increases the likelihood of hyperkalemia, especially in the elderly. The trimethoprim portion acts to prevent potassium excretion in the distal tubule of the nephron. Spironolactone has been reported to induce the enzymes CYP3A4 and certain UDP-glucuronosyltransferases , which can result in interactions with various medications. However, metabolites of spironolactone reportedly irreversibly inhibit CYP3A4. In any case, spironolactone has been found to reduce the bioavailability of oral estradiol, which could be due to induction of estradiol metabolism via CYP3A4. Spironolactone has also been found to inhibit UGT2B7.
The Russian Foreign Ministry summoned senior US diplomats after comments on 21 May by US National Security Adviser Jake Sullivan appeared to support Ukrainian attacks on Russian-annexed Crimea using Western weaponry. The German Defence Ministry said it was moving its Patriot missile battery deployed in Slovakia to Lithuania to secure the NATO summit to be held in Vilnius in July. The Prosecutor-General of Ukraine released figures showing that at least 483 children had been killed during the conflict and nearly 1,000 others were wounded, with the National Social Service of Ukraine saying that nearly 1,500 children had been orphaned. Most of these casualties came from Donetsk Oblast, with 462 recorded individuals. 278 casualties were recorded in Kharkiv Oblast, 128 in Kyiv Oblast, 102 in Kherson Oblast, 91 in Zaporizhzhia Oblast, 89 in Mykolaiv Oblast, 72 in Dnipropetrovsk Oblast, 70 in Chernihiv Oblast and 67 in Luhansk Oblast. UNICEF also said an estimated 1.5 million Ukrainian children were at risk of depression, anxiety, post-traumatic stress disorder and other mental health issues. The intelligence directorate of the Ukrainian Defence Ministry warned of a plot by Russia to stage an incident at the Zaporizhzhia Nuclear Power Plant to stall Ukraine's anticipated counteroffensive. Canada's Defence Minister Anita Anand announced that her country would supply the Ukrainian Air Force with 43 AIM-9X Block II Sidewinder missiles to help intercept Kh-101 and Shahed type Iranian made Drones. === Border reopening: August–September 2023 === On 22 August 2023, a flight from Pyongyang to Beijing arrived, marking the first known international commercial flight to depart from North Korea since the country's borders were closed in January 2020. Additionally, four flights between Pyongyang and Vladivostok, Russia, were scheduled for that month. On 27 August 2023, the Korean Central News Agency reported that North Korea would allow its citizens living abroad to return to the country for the first time since travel restrictions were imposed in early 2020. However, these returning citizens were still required to undergo a one-week quarantine. On 25 September 2023, North Korea lifted its travel ban on foreign nationals, as reported by China's CCTV. Foreigners would still be subject to a two-day quarantine upon their arrival.
==== Water industry ==== The water industry provides drinking water and wastewater services (including sewage treatment) to households and industry. Water supply facilities include water wells, cisterns for rainwater harvesting, water supply networks, and water purification facilities, water tanks, water towers, water pipes including old aqueducts. Atmospheric water generators are in development. Drinking water is often collected at springs, extracted from artificial borings (wells) in the ground, or pumped from lakes and rivers. Building more wells in adequate places is thus a possible way to produce more water, assuming the aquifers can supply an adequate flow. Other water sources include rainwater collection. Water may require purification for human consumption. This may involve the removal of undissolved substances, dissolved substances and harmful microbes. Popular methods are filtering with sand which only removes undissolved material, while chlorination and boiling kill harmful microbes. Distillation does all three functions. More advanced techniques exist, such as reverse osmosis. Desalination of abundant seawater is a more expensive solution used in coastal arid climates. The distribution of drinking water is done through municipal water systems, tanker delivery or as bottled water. Governments in many countries have programs to distribute water to the needy at no charge. Reducing usage by using drinking (potable) water only for human consumption is another option.
==== Complex IV ==== In Complex IV (cytochrome c oxidase; EC 7.1.1.9), sometimes called cytochrome AA3, four electrons are removed from four molecules of cytochrome c and transferred to molecular oxygen (O2) and four protons, producing two molecules of water. The complex contains coordinated copper ions and several heme groups. At the same time, eight protons are removed from the mitochondrial matrix (although only four are translocated across the membrane), contributing to the proton gradient. The exact details of proton pumping in Complex IV are still under study. Cyanide is an inhibitor of Complex IV.
== NMR spectroscopy on large proteins == Traditionally, nuclear magnetic resonance spectroscopy has been limited to relatively small proteins or protein domains. This is in part caused by problems resolving overlapping peaks in larger proteins, but this has been alleviated by the introduction of isotope labelling and multidimensional experiments. Another more serious problem is the fact that in large proteins the magnetization relaxes faster, which means there is less time to detect the signal. This in turn causes the peaks to become broader and weaker, and eventually disappear. Two techniques have been introduced to attenuate the relaxation: transverse relaxation optimized spectroscopy (TROSY) and deuteration of proteins. By using these techniques it has been possible to study proteins in complex with the 900 kDa chaperone GroES-GroEL.
